Yesterday I got 255 miles on half a tank. Trip computer reads 31mpg average. It was primarily highway driving at 75-80mph and some city mixed in.
In the past I've been getting 270 miles on a FULL tank. No clue what happened overnight...this car turned into a Hybrid or something

This is typical and happens, in my experience THREE times during the life of an Audi at the three following milage intervals.
Note: these are "in the spirit of" miles. It may happen earlier or later, but typically in the vicinity of the following.
5,000
10,000
15-20,000
Another kinda cool thing is that the POWER improves @ about these milage intervals, too.

Could be a number of factors for MPG to change overnight: outside temperature, tire pressure, etc... All things equal it's most likely the gas that you put in your car. Filling on 90/10 ethanol mix will cause a negative impact on MPG while filling on more pure gas mix will increase it. Even if you fill at the same gas station you may still be affected.

During summer I got over 36 MPG by using cruise control at about 60 MPH. Trip computer is optimistic by about 1 MPG but still impressive. Of course using cruise control is no fun, I typically get between 26 and 30 MPG on the highway.

Two weeks ago I took a trip to Florida from South Carolina in my 2.0T Quattro. 90% of the travel was on I-95. I had it on cruise control for most of the time and I got a little over 34 MPG on the interstate part of the trip.
