can we run a 245/45/18 tire or will there be rubbing issue..yes I know the speed will be off.

well that is factory...245/45/18 are taller tires and i just don't want to rub

I have been running 255/40/18s for 3 years without a problem.

the OD of your tire will increase by 24.5 mm. Just shy of an inch.

Default heres the answer
While I dont have the experience myself , I saw one with 245/45/18 at my sports club and asked the guy if he had problems, he said he doesnt, ( note that is not an S line)
I asked a wheel and tire salesman ( who knows audis very well) and told me that with S line or eibach springs, the higher he suggested is 255/40/18 , he said that 245/45/18 and a lowered car will have rubbing problems.
