I think it makes a difference. The S8 comes with it as standard and the sound
levels are much lower than on the A8. However, I cannot tell how much is due to the glass and how much comes from other features.
I believe the doors get different window motors and mechanisms to handle the extra weight and thickness

I've had an '04 without it and a '06 with it
Clearly worth the money - it makes a difference.
Another upgrade that was worth every penny was the wood steering wheel and shift ****. It makes me smile every time I get in the car. The old shift **** seemed so cheap and out of place compared to the quality of the rest of the interior.
