2008 S5 4.2 Ticking/Knocking

Finished swapping the motor on this 08 S5 4.2. Engine has 144K kilometres on it. Did not do major work, only gaskets, seals, clutch, spark plugs, etc. Got everything primed correctly, started up great and idled well. No faults, warmed up and took it for a drive down my street (no hard pulls). Suddenly the car started misfiring a bit and this ticking/knocking sound coming from driver side bank started (posted YT link above). Only Faults were hide cylinder and cyl 6 misfire. Checked timing values and it was to specification. Removed both valve covers to check for loose rockers, damage to the cams and removed both hpfp’s to inspect the buckets and rollers. Nothing looks damaged or out of place. I flushed and changed the oil once again running 5W-40 Liqui moly. Im stuck at the moment on what to do and where to start looking next. Any advice and tips would be greatly appreciated.
