I use that button all the time. Like Dave said, the ignition has to be on for it to work. I rewired mine with constant power because that was pissing me off. It is much more usable now. It should also be noted that if you trunk is in valet mode (key in trunk turned 90 degrees clockwise) the red button also will not work.
Troubleshooting should be easy, it is an "ignition on" powered circuit with a momentary contact switch that activates a single actuator. It is not tied into the central locking system at all. I am not sure which fuse runs it. I would check your trunk lock "status" and make sure you are using it with the ignition on.

I know that it won't if it is in valet mode. But never tried it when just locked.

wow! what a great guide, it told me everything i needed to know. My trunk hadn't worked on my audi since the day i bought it (obviously not brand new
). I had no idea where to even start, I thought i would need to buy a new trunk lock. But thanks to your guide my trunk is working again!
