I know I'm a jaded Audi guy. But I did take a few quick trips in the GLK.
Not a terrible car by any means. But there is an element of cheapness on the inside. The V6 was peppy. About the only thing I liked on it was the 20 inch wheels.
The radio display is a bit laughable on the GLK. The MMI interface on the Q5, even on a non-navigation car really makes the MB look low rent.
There's really no reason to buy the GLK over a Q5. Other other than you want to drive around in a Mercedes or just like the looks of the Mercedes better.

There was a great description in one of the automotive magazines a few years ago for a review of the prior generation C Class. I don't remember it exactly so I'm paraphrasing: The A4 (B6), inside and outside, feels like a destination, while the C Class feels like you're still on your way. The high quality finish and design of the A4 looks like you've arrived. You feel perfectly content to not upgrade to an A6 or A8. The C Class, on the other hand, feels like the entry level car that it is. You're constantly reminded of what you're missing out on in the E and S Classes. I test drove the C and have to agree with this. The look of the GLK reminds me of the C Coupe a few years ago, "Mercedes-Lite," which the NY Times calls "Mercedes playing the value card" (and not in a good way). The C Coupe died a quick death and time will tell if the GLK will meet a similar fate.

glk vs q5
The GLK does have a unique old school (and desirable) upright stance to the ergonomics. I miss the day of a nice shallow dash with high sight lines to the hood corners. The GLK has this in spades. From the outside the G wagen look does not translate as well but still the car looks good.
The GLK has a few other pluses. The V6 sounds great and is a world class proven design. Will run bullet proof for years and years making it a safer long term purchase. It is a gas pig though based on sized and weight.
The GLK is Mercedes. For some that means no further explanation required. Audi is not Mercedes. Not in status.
Other than that the Q5, is hands down to me the better value. Even the base 2.0T is wonderful coming standard with a great economical motor, leather memory seats, bluetooth and all the essential goodies save for sunroof and xenon (canadian base).
The Q5 is more spacious both passengers and cargo. That is a major plus as the GLK is really quite a small vehicle for the money.
The Q5 has Audi jewel like build quality and, imo, the nicest and friendliest interior set-up bar none. I find the mmi flawless in my A4 Avant.
Lets not forget the coup de grace....the Q5 has quattro. Nobody does awd like Audi, nobody....
