Unfortunately my news was quite bad. Audi have recommended to replace the Mechatronic Unit as I had suspected going by what I researched on forums.
At £2.5k for the unit alone, I feel like I've hit a brick wall! The car is immaculate and has years more driving so I've to bite the bullet and find the cash somehow.
I sourced an independent gearbox specialist locally who knew exactly what I was talking about as soon as I contacted him. "common fault in those cars" he said. He's done the job many times, but stated he only orders the new mechatronic unit from Audi Germany and services the gearbox at the same time, otherwise its pointless.

Well like most said it most likely is some faulty sensor somewhere.. all i was trying to tell u is that if u leave ure car parked on a hill or grade.. whether at work or at home it may eventually damage the box... the gear box oil may be settling more where its not meant to be... just keep an eye out that is all
Now this may be due to the fact I've recently just discovered I've got a faulty mechatronic unit, as a seal ruptured and fluids have made their way into the wiring loom of the Mechatronic.
But there may be some truth in this theory about gear oil settling somewhere it shouldn't be... Especially as my car sits in the same position every night, nose-down. The fact Audi advise on specific towing instructions also point to this theory??

For anyone that's interested - my mechatronic unit was sent to ECUTESTING.COM today to be reconditioned.
Will provide updates as soon as I have them. They give a 2-3 day turnaround, so fingers crossed for a quick repair..

Quiggs any update since sending in your mechatronic unit to ECUTESTING.COM?
I am new to this forum, and an owner of a 2013 S4 (B8) (128,000 miles).
I have also encountered the unfortunate situation where the DSG does not engage when accelerating/reversing slowly from a stop and then suddenly lurching forward after having been completely limp at the lower RPMs. Then my dashboard lights up, displaying DSG transmission failure, along with the TPMS lights.

I've done around 200 miles since getting mine back on the road.
ECUtesting didn't have to replace any parts of my mechatronic unit, but it did need cleaning. Due to the fact I took my car off the road immediately the very first time I got the dreaded gearbox malfunction message on the dash, I saved any further damage.
So repair of my wiring harnesses, new coolant pump and lots of lovely new gear oil with a cleaned Mechatronic Unit from ECUtesting and I'm back on the road a very happy man! All in, my repairs cost me a little shy of £900. Unbelievable as Audi wanted £2.7k alone for a new mechatronic unit, never mind labour @ £150 p/h and whatever else I'd be stung for. Gear Oil, wiring etc.
Definitely glad I took the advice of the Audi Dealership initially and drove the car no further - even though I contemplated it as the car was driving without issues.
Gear changes are definitely even smoother now than ever. And with a full engine service at the same time, the car is absolutely fabulous to drive.
I wish the other guys on here all the best in resolving their issues and thanks to everyone for the help/feedback.

I've done around 200 miles since getting mine back on the road.
ECUtesting didn't have to replace any parts of my mechatronic unit, but it did need cleaning. Due to the fact I took my car off the road immediately the very first time I got the dreaded gearbox malfunction message on the dash, I saved any further damage.
So repair of my wiring harnesses, new coolant pump and lots of lovely new gear oil with a cleaned Mechatronic Unit from ECUtesting and I'm back on the road a very happy man!
