Tip for city/commutin, 6speed for all else!
I personally got a tip, cause I live in miami, and have 2 hours of driving a day to and from work. The last thing I want to do after a double shift is slogging home and shifting back and forth every time some retard on a cell phone hits the brakes. The drivers down here are the dumbest this side of Istanbul. But when I get back up to New Hampshire, I'll be wishing I could be heeling and toeing through those empty hilly roads in a 6 speed.
The 6 speed is probably more fun just driving around, but the tip is the best automatic transmission out there (besides the F1 trannies out there). It is still pretty sporty in manual mode, and I love being able to kick it into 2nd when I want to, and then dump it back into drive when the traffic piles up.
In the end, it's all about how you plan to drive it, and how much you value that extra sporty feeling over the relaxation of a fantastic auto.

I have owned both.
Tip you just don't have to think about. Foot down, car fast. That simple.
I love my 6-speed, and I would do it again for certain, but the tranny in my car was defective, and it took a three month fiasco to get it replaced. I also had clutch bearing issues, and other people have synchro issues, so as nice as the 6-speed is, its one more thing to worry about.
One thing that is most often left out in these discussions is that the tip feels a LOT faster around town. From a stop in any sort of traffic (where you're not going to be hitting 50-70 mph), a tip is going to take a 6-speed off the line. The S4 manual tranny just can't take being beaten on, and the combination of that and the pitifully low 1st gear makes for very slow "normal" launches.
Of course, I do LOVE the sound my Borla makes every time I go to shift the 6-speed.

The tiptronic is more sophisticated for city driving:
you get to keep both hands on the wheel and can concentrate on breaks in traffic and other cars rather than rowing the boat. Also, if your commute involves any traffic congestion, the tip makes for a more relaxed ride.

Tip is sweet... with F1 style steering wheel shifting...
it's a blast to drive fast! (don't let the old rowing folks tell you otherwise) Hell, when everyone first heard about the tip on the Porsche, it was big news... new technology.. the future...

As long as the trans has a torque converter, it's not a true "clutchless 5 speed manual" in my book
BMW's SMG setup or the F1 trans option on some Ferrari's is what Audi should be developing for a future "S" car, IMHO.
